With the continuous development of microscope research and development technology, the types of microscopes are now very rich, and the resolution is also constantly improved. Since the beginning of the 21st century, a variety of ultra-high-resolution fluorescence imaging technologies have emerged one after another, breaking the limit of the resolution of optical microscopes and increasing the resolution to the scale of tens of nanometers. However, it is still a huge challenge to further improve the resolution of the optical microscope to the molecular level to observe the nano-scale subcellular structure and even the structure within a single biological macromolecule. In response to this phenomenon, Chinese scientists have developed a new microscope technology.
According to news from the Ministry of Science and Technology, Chinese scientists have developed a new interference single-molecule localization microscope technology, which is also called Repetitive Optical Selective Exposure (ROSE), by using rapidly modulated structured light. It is known that the intensity of fluorescent molecules is closely related to the phase of the interference fringe. The interference single-molecule positioning microscope uses six different directions and phase interference fringe to excite the fluorescent molecules. The precise relationship between the intensity of the fluorescent molecules and the phase of the interference fringe can be used to determine the accuracy of the fluorescent molecules location information. The interference single-molecule localization microscope can resolve DNA origami arrays with a point distance of 5 nm. The resolution of the microscope is improved to a molecular scale within 3 nm. The accuracy of single-molecule positioning is close to 1 nm. The theoretical positioning accuracy is traditional 2.4 times.
In summary, interference single-molecule localization microscopes provide a more powerful means of observation for life science research, further breaking the existing limits of optical microscope resolution. Related research results were published in the journal Nature Methods.
